The riverfront trail is a pretty good one. The trail winds through some industrial areas but one you get a couple of miles north of downtown, the trail is quite nice. I'd avoid going alone if possible. The trail goes next to a couple of questionable neighborhoods. The view of the river is unbeatable and the crossing at the Chain of Rocks bridge is awesome. If you fee really good, you can keep going all the way up to Alton. That's a pretty good ride but very scenic.
if urban biking is more your style, youmight like to check out http://www.bikestlouis.org/ , a trail which opened officially in april. if you're interested in a riding partner, i'm still a noob to urban biking and wouldn't mind some company (ie a second set of eyes)

I can recommend the river des peres trail. It is relatively flat, there are nice pedestrian stops at Morgan Ford, Germania and River Des Peres. It's a fun ride, but it's short if you're trying to get exercise, you may have to do several runs. But, you can start at Gravois/River Des Peres and ride to Holly Hills Blvd. to Carondelet Park and ride around their pedestrian trail.
I still haven't ridden the riverfront trail. If you have any experience that one, let me know.
